    SummaryViral RNA-dependent RNA polymerases (RdRp) differ from DNA-dependent RNA polymerases, DNA-dependent DNA polymerases, and reverse transcriptases in that RdRps contain “fingertips” consisting of several polypeptide strands in the fingers domain interacting with the thumb domain. The crystal structure of bovine viral diarrhea virus (BVDV) RdRp containing an Asn438 duplication shows that the “N-terminal domain,” which occurs only in pestiviruses such as BVDV, interacts with the polymerase component of the same polypeptide chain. This contrasts with the domain swapping observed in the previously determined structure of the BVDV NADL strain RdRp. By comparison with the NADL structure and through the use of biochemical data, it is possible that the N-terminal domain, in conjunction with the fingertips, is required to bind and assist the translocation of the RNA template. The partial disorder of the loop containing the additional Asn438 residue may explain the low replication rate of the recombinant compared with the wild-type virus

    3D Arbitrary Channel Fabrication for Lab on a Chip Applications using Chemical Decomposition

    This article demonstrate a simple method to use of three-dimensionally (3D) printed molds that are chemically decomposable for rapid fabrication of complex and arbitrary microchannel geometries. These complex microchannel are unachievable through existing soft lithography techniques. The molds are printed directly from hand held 3D printing pen that can print in midair, making rapid prototyping of microfluidic devices possible in hours. PLA based copper filament is used to print the arbitrary channels. The printed channels are then placed inside PDMS and PDMS is cured. The cured sample is then immersed in chemical solution (Acetic Acid + Sodium Chloride+ Hydrogen peroxide), which decomposes the PLA based copper channel thus leaving an empty channel inside the PDMS block. This method enable precise control of various device geometries, such as the profile of the channel cross-section and variable channel diameters in a single device

    Dispersion Curve Engineering of TiO2/Silver Hybrid Substrates for Enhanced Surface Plasmon Resonance Detection

    As surface plasmon resonance (SPR)-based biosensors are well translated into biological, chemical, environmental, and clinical fields, it is critical to further realize stable and sustainable systems, avoiding oxidation susceptibility of metal films—in particular, silver substrates. We report an enhanced SPR detection performance by incorporating a TiO2 layer on top of a thin silver film. A uniform TiO2 film fabricated by electron beam evaporation at room temperature is an effective alternative in bypassing oxidation of a silver film. Based on our finding that the sensor sensitivity is strongly correlated with the slope of dispersion curves, SPR sensing results obtained by parylene film deposition shows that TiO2/silver hybrid substrates provide notable sensitivity improvement compared to a conventional bare silver film, which confirms the possibility of engineering the dispersion characteristic according to the incidence wavelength. The reported SPR structures with TiO2 films enhance the sensitivity significantly in water and air environments and its overall qualitative trend in sensitivity improvement is consistent with numerical simulations. Thus, we expect that our approach can extend the applicability of TiO2-mediated SPR biosensors to highly sensitive detection for biomolecular binding events of low concentrations, while serving a practical and reliable biosensing platform

    Verifying multi-partite mode entanglement of W states

    We construct a method for verifying mode entanglement of N-mode W states. The ideal W state contains exactly one excitation symmetrically shared between N modes, but our method takes the existence of higher numbers of excitations into account, as well as the vacuum state and other deviations from the ideal state. Moreover, our method distinguishes between full N-party entanglement and states with M-party entanglement with M<N, including mixtures of the latter. We specialize to the case N=4 for illustrative purposes. In the optical case, where excitations are photons, our method can be implemented using linear optics.Comment: 11 pages, 12 figure

    Higgs Structures of Dyonic Instantons

    We study Higgs field configurations of dyonic instantons in spontaneously broken (4+1)-dimensional Yang-Mills theory. The adjoint scalar field solutions to the covariant Laplace equation in the ADHM instanton background are constructed in general noncanonical basis, and they are used to study explicitly the Higgs field configurations of dyonic instantons when the gauge fields are taken by Jackiw-Nohl-Rebbi instanton solutions. For these solutions corresponding to small instanton number we then consider in some detail the zero locus of the Higgs field, which describes the cross section of supertubes connecting parallel D4-branes in string theory. Also the information on the Higgs zeroes is used to discuss the residual gauge freedom concerning the Jackiw-Nohl-Rebbi solutions.Comment: 1+27 pages, 6 figure
